Generation and characterization of a high‐affinity chimeric anti‐OX40 antibody with potent antitumor activity

OX40 is a costimulatory molecule that belongs to the tumor necrosis factor receptor (TNFR) superfamily. agonist-based combinations are emerging as promising candidates for novel cancer immunotherapy. Clinical trials have shown agonist antibodies could lead better results in patients. Using hybridoma platform and three different types of immunization strategies, namely recombinant protein, DNA, overexpressing cells, we identified chimeric anti-OX40 antibody (mAb035-hIgG1 from DNA immunization) shows excellent binding specificity, slightly stronger activation human memory CD4+ T cells similar potent antitumor activity compared with BMS 986178, an currently being evaluated treatment solid tumors. This paper further systematically investigates antigen-specific immune response, number binders, epitope bins, functional activities among strategies. Interestingly, found strategies affect biological monoclonal antibodies.
